人XIII因子[EC=2.3.2.13]
因子XIII是谷氨酰胺肽g-谷氨酰胺转移酶因子XIIIa(纤维蛋白连接酶、血浆转谷氨酰胺酶、纤维蛋白稳定因子,E.C. 2.3.2.13)的酶源形式(1-3)。因子XIII在转氨酶中是独一无二的,因为它在体内是一种酶原(2)。因子XIII既存在于细胞外的血浆中,也存在于细胞内的血小板、巨核细胞、单核细胞、胎盘、子宫、肝脏和前列腺组织中。血浆中的XIII因子在肝脏中合成,并以四聚体(Mr 20,000)的形式循环,由2对非相同的亚单位(A2B2)组成(4)。细胞内的形式是在组织中合成的,在那里它们是由2条相同的A链(A2)组成的二聚体(Mr 6000)(7-11)。血浆和细胞内形式的因子XIII的A亚单位在功能上是相同的。A亚单位含有6个自由巯基,其中一个是活性部位(12)。
血浆中XIII因子(A2B2)的浓度约为30mg/ml(8)。它是凝血级联中最后一个被激活的酶原,是该系统中唯一不是丝氨酸蛋白酶的酶。血浆因子XIII(A2B2)转化为活性转氨酶因子XIIIa(A2')的原因是凝血酶水解了A亚基NH2末端的Arg36-Gly37(13)。只有在B亚基二聚体与A2'二聚体发生Ca2+(Kd-3M)和纤维蛋白(Ogen)(Kd-8M)依赖性解离后,活性才得以完全表达(14-16)。

Storage and Stability
May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
