有多种酶可以水解 ATP 或 GTP,形成 ADP 或 GDP 和无机磷酸盐 (Pi)。我们的酶联无机磷酸盐测定法(ELIPA)可以实时测量水解过程中释放的磷酸盐。这样就能为酶的活性提供一种动力学检测技术。适用于这种分析的纯化酶包括信号磷酸酶、磷酸酶、驱动蛋白马达(见图 1)、代谢酶、膜转运体和碱性磷酸酶。一般来说,这种检测方法适用于 Kcat 值高于 0.1 的酶,如果您的酶或酶家族的 Kcat 值较低,那么 CytoPhosTM 磷酸盐检测试剂盒(订货号:BK054)就很有用。如果酶的活性很低,如小 G 蛋白,非放射性 GAP 分析法(Cat. # BK105)或放射性替代方法 EasyRad 磷酸盐分析法(Cat. # BK055)是理想的选择。
