SIRT1荧光分析试剂盒是一套完整的检测系统,用于测量 Sirtuin 1 (SIRT1) 的活性,以进行筛选和分析应用。它采用方便的 96 孔格式,包含 100 次荧光 SIRT1 (Sir2) 活性测定所需的全部试剂。此外,试剂盒还包括纯化的 SIRT1 酶和 SIRT 抑制剂烟酰胺,分别用作阳性和阴性对照。SIRT1荧光分析试剂盒基于独特的荧光底物和显影剂组合。这种检测方法无需处理传统检测方法中的放射性、提取和层析问题。使用该试剂盒,只需在微孔板上进行两个简单的步骤即可分析 SIRT1 活性水平。首先,将含有乙酰化赖氨酸侧链的 HDAC 荧光底物(HDAC 底物 1)与纯化的 SIRT1 酶孵育。脱乙酰化会使底物敏化,因此随后用赖氨酸显影剂处理会产生荧光团,然后可以用荧光分析仪进行测量。SIRT1荧光分析试剂盒采用便捷的 96 孔格式,内含试剂、纯化的 SIRT1 酶和 SIRT 抑制剂烟酰胺,可用作 100 次荧光 SIRT1 (Sir2) 活性测量所需的阳性和阴性对照。
