肽基-脯氨酰异构酶（PPIase） Pin1 可催化磷酸丝氨酸/苏氨酸-脯氨酸肽键的顺式/反式异构化。Pin1 由较短的 N 端蛋白-蛋白相互作用结构域和较长的 C 端izomerase 结构域组成，前者可使酶与磷蛋白结合。Pin1 有许多生物底物，在细胞周期调控中发挥关键作用，并在许多人类癌症中上调。最近，Pin1 与阿尔茨海默病的发病机制有关。Pin1 可与 Tau 蛋白的 Thr-212/231 磷酸化残基结合，增加其去磷酸化，从而防止 Tau 神经纤维缠结的形成。
绿色荧光PIN1活性分析试剂盒使用经过预处理以转化为顺式异构体的荧光底物。Pin1 将底物转变为反式构象，反式构象很容易被裂解，从而产生荧光信号，可在 Ex/Em=490/520nm 波长下监测荧光信号。荧光强度的增加与 Pin1 的活性成正比。
