PARP1检测试剂盒是一种荧光偏振（FP）均相96孔或384孔检测试剂盒，旨在以高通量筛选（HTS）形式筛选和分析能将PARP2捕获到DNA上并形成复合物的小分子。PARPtrap™ PARP1检测试剂盒采用便捷的96孔和384孔形式，配备纯化的重组PARP2酶（氨基酸2 - 583）、荧光标记的寡核苷酸双链体、NAD+ 以及用于100次或400次酶反应的PARPtrap™ 检测缓冲液。
该荧光探针是一种可在溶液中自由旋转的小分子，因此荧光偏振（FP）较低。在无核糖基化作用时，PARP1与荧光探针结合，形成大型复合物，从而发射出高偏振光。在自核糖基化步骤之后，PARP1从荧光寡核苷酸双链体上解离，荧光探针随后可再次自由旋转（低FP）。在存在PARP1抑制剂的情况下，PARP1会与荧光寡核苷酸双链体结合，产生高FP。FP信号的增加与PARP结合量成正比。
