UbcH6(也称为泛素结合酶E2 E1,UBE2E1)是一种E2泛素结合蛋白,它以ATP依赖的方式从泛素激活酶(E1)接收泛素,并将其转移至E3连接酶。UbcH6受泛素特异性蛋白酶7(USP7)调控,且是PRC1(多梳抑制复合体1)的组成部分,PRC1是一种能使组蛋白H2泛素化的E3复合体,因此参与干细胞和癌细胞的增殖过程。UbcH6还能与ataxin-1相互作用并使其泛素化,从而调节其活性。ataxin-1与脊髓小脑共济失调1型(SCA1)相关,这是一种常染色体显性遗传性神经退行性疾病。靶向UbcH6可能在癌症和SCA1的治疗中具有潜在的治疗价值。

Assay Conditions
50 µl reaction mix (4 mM ATP, 5 mM MgCl2, 1 mM DTT, UBE1 as an E1 (0.5 µg), UbcH6, Biotin-Ubiquitin (0.5 µg) and Smurf2 as an E3(2 µg) in 50 mM Tris pH 7.4) for 30 min at 37°C, then add to streptavidin coated plates for 30 min. at 37°C. Incubate with anti-Smurf2, then Europium-labeled secondary antibody, and detect by time resolved fluorescence (λex=330 nm, λem=620 nm).
Instructions for Use
Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
Applications
Useful in conjunction with E1 and E3 for the study of enzyme kinetics, screening inhibitors, and selectivity profiling.
