抗生素嘌呤霉素是氨酰-tRNA的结构类似物,由于它能特异性地掺入新生多肽链的C端从而终止翻译,长期以来一直被用作蛋白质合成的抑制剂。基于这一特性,一种全新且速度显著更快的蛋白质标记方法已被开发出来:

在低浓度标记的脱氧胞苷-嘌呤霉素结合物存在的条件下,可通过体外翻译制备C端标记的全长蛋白[1]。
这些蛋白质已成功用于分析蛋白质-蛋白质相互作用(下拉实验、荧光相关光谱、蛋白质-蛋白质微阵列)和蛋白质-DNA相互作用(DNA微阵列)[2,3,4]。
与传统的翻译后标记方法相比,由于蛋白质表达与标记的同步进行,可避免耗时的额外纯化步骤。
