保护素D1是一种由二十二碳六烯酸(DHA;产品编号90310)合成的特殊促消退介质(SPM)。1 DHA被氧化为16S,17S-环氧保护素,后者经酶促反应转化为保护素D1。保护素D1能以非促炎方式增强巨噬细胞对凋亡多形核白细胞(PMN)的吞噬作用,且在巨噬细胞与凋亡细胞相互作用过程中于体外产生。2 它在24小时后可增强小鼠的吞噬作用,但在炎症起始或高峰期则无此作用。在酵母聚糖诱导的小鼠炎症模型中,每只小鼠给予300 ng的保护素D1,可减少PMN的浸润。在气管内脂多糖(LPS)滴注诱导的小鼠肺损伤和炎症模型中,保护素D1(200 µg,静脉注射)可抑制支气管肺泡灌洗液(BALF)中中性粒细胞计数的增加以及肺髓过氧化物酶活性。3 它还能减轻肺水肿,并促进支气管肺泡灌洗液中中性粒细胞的凋亡。
