组蛋白是真核细胞染色体的蛋白质部分的主要成分。它们富含氨基酸精氨酸和赖氨酸，并在进化过程中得到了极大的保守。组蛋白将DNA包装成染色质的紧密团块。每类H2A、H2B、H3和H4的两个核心组蛋白组合起来，被146个碱基对的DNA包裹，形成一个八聚体核糖体。组蛋白尾部经过许多翻译后修饰，直接或间接地改变染色质结构，以促进转录激活或抑制或其他核过程。除了遗传密码外，不同组蛋白修饰的组合显示了所谓的 "组蛋白密码"。组蛋白甲基化和去甲基化分别由组蛋白甲基转移酶和组蛋白去甲基化酶动态调节。组蛋白变体H2A.Z与活性基因有关。
