HER4(人表皮生长因子受体4),也被称为ErbB4或受体酪氨酸激酶4,是EGFR家族的一种受体酪氨酸激酶。它是心脏、神经系统和乳腺发育所必需的物质,也存在于神经肌肉接头的突触后侧和表皮中。HER4可被神经调节蛋白2-4、β细胞素以及肝素结合性表皮生长因子样生长因子(HB-EGF)激活,并参与细胞有丝分裂和细胞分化过程。HER4与配体结合后会发生构象变化,与其他HER蛋白形成同二聚体或异二聚体,进而激活丝裂原活化蛋白激酶(MAPK)通路和磷脂酰肌醇3-激酶(PI3K)/AKT通路。该蛋白的突变可能引发癌症,同时也与19型肌萎缩侧索硬化症(ALS)和精神分裂症相关。该蛋白作为癌基因或抑癌基因的作用尚未完全明确,可能取决于癌症类型。目前已开发出多种HER4抑制剂并正在开展临床试验。阿法替尼是一种泛HER抑制剂,已获批用于治疗非小细胞肺癌(NSCLC),但可能导致急性肝损伤甚至死亡。研发针对HER4的高特异性抑制剂有望在肿瘤学领域发挥重要作用。
重组人HER4(人表皮生长因子受体4,又称ErbB4),包含26-651位氨基酸(胞外结构域)。该构建体在C端融合了人IgG1的Fc结构域,其后连接一个C端Avi-Tag™标签。该蛋白经亲和纯化获得。

Assay Conditions
The NRG1β Protein, Human (HEK293) was coated onto a 96-well plate overnight at 4°C (50 µl/well at a concentration of 0.5 µg/ml in PBS). The plate was washed 3 times with PBST and blocked using 200 µl of Blocking Buffer 3 for 2 hours at Room Temperature (RT). After removing the blocking buffer, 50 µl/well of purified HER4, serially diluted in 1x PP-02 Buffer, was added for 1 hour at RT. After washing, the plate was incubated for 40 minutes with HRP-labeled anti-FC antibody diluted 1:1000 in Blocking Buffer 3 (50 µl/well) and washed again. Finally, 100 µl of ECL substrate was added to each well and plate was read immediately in a luminometer or microtiter-plate reader capable of reading chemiluminescence.
Instructions for Use
Thaw on ice and gently mix prior to use. DO NOT VORTEX. Perform a quick spin before opening. Aliquot into small volumes and flash freeze for long term storage. Avoid multiple freeze/thaw cycles.
Applications
Useful for studying the binding of HER4 in ELISA and in cellular assays.
