p70S6K 负责 40S 核糖体蛋白 S6 的磷酸化，在人类成人组织中广泛表达。在完整细胞中，p70S6K 可被血清激活，且该激活过程可被渥曼青霉素和雷帕霉素抑制。p70S6K 的活性具有细胞周期依赖性，在从 G0 期释放的 G1 期细胞中，其活性可提升 20 倍。p70S6K 的激活需要对推定的自抑制假底物结构域中的丝氨酸残基、被 mTOR 磷酸化的苏氨酸 389 以及被磷酸肌醇依赖性激酶 1（PDK-1）磷酸化的苏氨酸 229 进行依次磷酸化。
